Experimental Induction of Leukaemia in Mice by Benzene and The Effect of Garlic (Allium Sativum) in Protection and Treatment

This experiment was designed for induction of leukaemia in mice by S/C injection of benzene in order to study the pathogenesis, blood morphology and effect of garlic treatment.
257 mice were divided into 5 groups:
1. Normal control group of 15 mice (10 females and 5 male) for normal blood parameters.
2. Control group for garlic extract of 15 mice (10 females and 5 males) was injected by 400 mg/kg b. wt garlic extract by intraperitoneal injection.
3. Group for olive oil of 15 mice (10 females and 5 males) was injected 0.1 ml olive oil S/C.
4. Benzene toxicity group 106 mice were injected by 0.001 ml benzene and o.1 ml olive oil S/C, the dosing continued once weekly for 9 monthes.
5. Benzene toxicity with garlic extract group 106 mice was injected by 0.001 ml benzene and 0.1ml olive oil and in the same time injected wit 400 mg/ kg b. wt garlic extract by i/p injection, the dosing continued once weekly for 9 monthes.
Benzene toxicity caused normocytic normochromic anemia as indicated by the indices of RBCs number, size and Hgb content. The character of anemia changed from microcytic hypochromic until the end of the experiment.
The index of MCHC gave a more detailed information on the character of anemia i.e from the 3rd week to the 12th week microcytic, then from the 12th week to the 15th week acrocytic and from the 15th week to the 21th week again became microcytic, it was hypochromic from the 3rd week to the 21th week.
Benzene toxicity induced the appearance of various types of poikylocytosis together in the same animals including:
1. leptocytes
2. basophilic stippling
3. polychromatic cells
4. spherocytes
5. stomatocytes
6. target cells
7. bar cells “Manar cells”
8. tear drop cells
9. rouleaux formation and echinocytes.
Garlic treatment was very effective on the following:
1. Normalized the RBCs count at the 9th week until the 12th week.
2. Gave protection against the formation of basophilic stippling.
3. Prevented the formation of rouleaux completely
4. Normalized and elevated the platelets number in few weeks.
Garlic treatment corrected but not completely the following:-
• Started by increase Hgb content only after the 24th week.
• PCV confirmed the little increased in Hgb content from the 21thweek to the 30th week, and from the 34th week to the 39th week
• Has a little effect on the number of polychromatic cells.
• Slightly corrected the number of spherocytes.
• Offered little correction for the number of stomatocytes only after the 27th week.
• Gave little protection against the target cells after the 24th week.
• Caused decrease of bar cells from the 24th week to the 30th week.
• Gave protection against tear drop cells until the 15th week.
• Gave protection against echinocytes from the 18th week to be normal to the end of the experiment.
Garlic treatment was completely non effective on the following:the appearance of leptocytes, mean corpuscular volume, mean corpuscular hemoglobin, mean corpuscular hemoglobin concentration.
Benzene toxicity caused the appearance of various types of myeloid leukaemia in the majority of mice. The cells of chronic myeloid leukaemia, neutrophilic metamyelocytes (0.3%) and neutrophilic band cells (0.3%). The cells of eosinophilic myeloid leukaemia, eosinophilic myelocytes (0.2%), eosinophilic metamyelocytes (0.3%) and eosinophilic band cells (0.2%).The cells of basophilic myeloid leukaemia, basophilic myelocytes (0.2%), basophilic metamyelocytes (0.1%) and basophilic band cells (0.2%).
Benzene toxicity caused the appearance of monoblastic leukaemia, monoblast cells (0.2%) and promonocytes (0.2%).
Garlic treatment was very effective on the following:
• Corrected the neutrophilic myelocyte to the the control level.
• Decreased the number of neutrophilic metamyelocyte below the number induced by benzene toxicity.
• Decreased the number of neutrophilic band cells below the number induced by benzene toxicity.
• Corrected the number of mature neutrophils to the control level below the number induced by benzenetoxicity.
• Corrected eosinophilic myelocytes to the control level, below that induced by benzene toxicity.
• Decreased the number of eosinophilic metamyelocytes below the number induced by benzene toxicity.
• Corrected number of eosinophilic band cells only three weeks during the experiment.
• Decreased the number of mature eosinophil cells below that induced by benzene toxicity.
• Garlic has no effect on the myeloblasts and the promyelocytes.
• Garlic has no effect on monocytic series.
